Overview of Centrosome-targeting Drugs
Centrosome is of important functions in regulating cell cycle, which makes it an attractive target for cancer treatment. Many compounds are used to against proteins involved in the regulation of different cell cycle points including drugs targeting the microtubule dynamics, Plks, Cdks, and AURK inhibitors. Drugs targeting microtubule dynamics include taxanes (paclitaxel and docetaxel) and vinca alkaloids (vinblastine, vincristine, and vinorelbine), which have been applied successfully for treating varieties of cancers, such as breast cancer, ovarian cancer and lung cancer. And a series of acridinones compounds have been found to be promising therapies against cancer causing G2-M arrest, inhibition of cell migration, and apoptosis.
